چکیده

Leukaemia is a heterogeneous hematologic malignancy characterized by unregulated proliferation of the early blood-forming cells which starts in bone marrow. The basic strategy leukaemia therapy involves induction leukemic apoptosis. Research on natural products have shown that some plant derivatives anticancer properties inducing apoptosis cells. Plants such as Camellia sinensis and Phyllanthus amarus are those had gained wide interest due to their anti-cancer effect. aim this study was investigate effects C. P. extracts human cell lines analysing cycle determining apoptotic state. were treated with ethanolic at concentrations 31.25 - 500 µg/mL for 24 h followed MTT assay determine IC50. IC50 U937 170±10.39 210±6.78 µg/mL, respectively. Flow cytometric analysis using Annexin V/propidium iodide (PI) staining also performed. extract 170 significantly increase U-937 (p<0.001), Jurkat (p<0.05) K-562 (p<0.01) when compared untreated Meanwhile, 210 induced both K562 but not caused arrest S phase (p<0.001) G0/G1 K652 control. Based findings, showed potential lines. In addition, has capability disrupt cycle.

Effects of berberine on proliferation, cell cycle distribution and apoptosis of human breast cancer T47D and MCF7 cell lines

Objective(s):Berberine, a naturally occurring isoquinoline alkaloid, has shown antitumor properties in some in vitro systems. But the effect of berberine on breast cancer has not yet been completely studied. In this study, we evaluated anticancer properties of berberine in comparison to doxorubicin. Materials and Methods: The antiproliferative effects of berberine and doxorubicin alone and in ...

Cell cycle arrest and apoptosis induced by oxaliplatin (L-OHP) on four human cancer cell lines.

BACKGROUND The effects of oxaliplatin (L-OHP) on cell cycle and apoptosis were examined. MATERIALS AND METHODS The HT29, MCF7, Hela and A549 cell lines were treated and dual parameter flow cytometry BrdU/PI was then performed to monitor cell cycle modifications. Annexin V-FITC and PI probes were used for the detection of apoptosis. RESULTS The cells were treated for 3 h followed or not by 2...

Gastroprotective and antioxidant activities of Phyllanthus amarus extracts on absolute ethanol- induced ulcer in albino rats

This study was designed to evaluate the gastroprotective and antioxidant effects of aqueous and acetone extracts of Phyllanthus amarus leaves in albino rats. P. amarus extracts (500 and 1000 mg/Kg) as well as cimetidine (100 mg/Kg) was administered orally once a day for two weeks before challenge with absolute ethanol (1 ml/ 200 g body wt). Pretreatment with P. amarus aqueous extract (500 mg/Kg...

Cell cycle kinase inhibitor expression and hypoxia-induced cell cycle arrest in human cancer cell lines.

Flow cytometric analysis of fibroblasts, normal breast epithelial cells and breast or other cancer cell lines identified variation in the abilities of cell lines to undergo cell cycle arrest as a response to hypoxia. Human mammary epithelial cells (HMEC), normal fibroblasts (Hs68 and WI38), HeLa cervical carcinoma and HTB-30 breast carcinoma cells arrest in G(1)/S in response to severe hypoxia....

Securinine from Phyllanthus glaucus Induces Cell Cycle Arrest and Apoptosis in Human Cervical Cancer HeLa Cells

BACKGROUND The Securinega-type alkaloids occur in plants belonging to Euphorbiaceae family. One of the most widely distributed alkaloid of this group is securinine, which was identified next to allosecurinine in Phyllanthus glaucus (leafflower). Recently, some Securinega-type alkaloids have paid attention to its antiproliferative potency towards different cancer cells. However, the cytotoxic pr...
